Influence of makroelements from food on DNA and epigenetic profile

The macroelements contained in food have an important function for the human body. They are involved in several of biochemical reactions in the body and their abundance can prevent serious diseases. The theoretical part of the bachelor thesis describes the function of minerals in the human body, the function of DNA and epigenetic mechanisms such as DNA methylation or histone modification. The influence of nutrition and function of selected macroelements – sodium, magnesium, calcium, potassium on epigenetic modifications and on the stability of G-quadruplexes was described. The aim of the experimental part of this work was to study the effect of these substances on DNA structures in vitro and to prepare them experimentally for in vivo studies.
Balance of energy, water and nutrients in the aquaponic cycle

Předložená diplomová práce byla zpracována s cílem vytvořit přehled poznatků v oblasti akvaponické potravinové produkce. Informace získané během tvorby tohoto přehledu pak měly vést, v kombinaci s daty získanými z funkčního provozu, k vytvoření matematického modelu akvaponického cyklu. Na akvaponické farmě provozované společností Flenexa plus s.r.o., která byla zdrojem potřebných procesních dat, měla být dále zpracována a vyhodnocena bilance energie a vody. Nakonec měla být v průběhu práce posouzena možnost implementace mikrořasového fotobioreaktoru do akvaponického cyklu. Úvod práce představuje motivaci vedoucí k potřebě inovovat dnešní potravinovou produkci. Kriticky jsou zhodnoceny predikce vývoje lidské populace, a to pak hlavně z pohledu dopadu, který by tento růst měl na zemědělskou produkci. Současná situace se na základě získaných poznatků ukazuje jako neudržitelná, primárně pak v oblastech vodohospodářství a energetické spotřeby. Následně je jako možné řešení vedoucí ke zlepšení udržitelnosti potravinové produkce zkoumána akvaponie. Akvaponie je definována a její jednotlivé komponenty jsou představeny z hlediska mechanismu jejich fungování a z pohledu jejich návrhu. Mezi popsané oblasti patří například principy tzv. coupled a decoupled akvaponie a popis možných typů hydroponického komponentu. V této části práce je pozornost věnována také představení cyklů jednotlivých živin v rámci akvaponie. Následující a poslední teoretická část práce je pak věnována mikrořasovému fotobioreaktoru. Jsou zde popsány mechanismy, jak motivující, tak odrazující od zakomponování bioreaktoru do akvaponie. V oblasti výhod se jedná hlavně o jeho roli ve stabilizaci pH a spotřebě toxikého amoniaku. Na druhou stranu jeho ekonomické dopady na profitabilitu akvaponie jsou velmi proměnlivé v závislosti na způsobu implementace. Samotný mikrořasový fotobioreaktor je pak v práci detailněji představen. Jednotlivé procesní ukazatele ovlivňující růst řas jsou rozebrány, a to společně s jednotlivými typy fotobioreaktoru, metodami sklizně a využitími pro vyprodukované mikrořasy. Na základě poznatků schromážděných v této práci pak lze jako nejvhodnější k implementaci do akvaponie doporučit hybridní fotobioreaktory, u kterých je většina osvětlení zajištěna v podobě slunečního svitu. Samotná experimentální část práce pak začíná popisem zkoumaného provozu společnosti Flenexa plus s.r.o. z pohledu aplikovaného akvaponického procesu. Jednotka podrobená měření byla provozně stabilní a využívala implementace hydroponického komponentu typu Deep Water Culture (DWC). Spolu s detailním popisem celého provozu jsou poskytnuty a vyhodnoceny vypracované bilance vody a energií. Pozornost je pak přesunuta k matematickým modelům vypracovaným a ověřeným na základě dat a poznatků shromážděných z provozu společnosti Felenexa plus s.r.o. Logika a algoritmy, na kterých jsou oba modely postaveny, jsou v této části vysvětleny a diskutovány společně s hlavními funkcemi a schopnostmi obou modelů. První, primárně statistický model je představen jako nástroj pro použití při uvádění akvaponie do provozu. Druhý, fyzikální model pak v uživatelsky přívětivém formátu představuje základ pro model řízení akvaponické farmy s mikrořasovým fotobioreaktorem. V neposlední řadě jsou nastíněny také cesty možného budoucího vývoje pro oba vytvořené modely. Práce je následně završena shrnutím a diskusí nad poznatky a výstupy získanými během celého tvůrčího procesu.
Changes in rainfall-runoff regime and biogeochemistry in headwater areas

Headwater areas are a very important current or potential drinking water source. Their ecological value for the whole ecosystem is equally important. Headwater areas are sites with minimal anthropogenic intervention and usually the least polluted water sources. Changes in climate and rainfall-runoff (R-R) regime together with water quality in these areas affect the status and availability of water further downstream significantly. It is also one of the most sensitive areas and is an early indicator of ongoing changes. This thesis focuses on the main issues that are already ongoing and are affecting the availability of drinking water significantly. Some of these are trends in R-R regime in the context of the climate change and glacier retreat in headwater areas. As a very good example of a mid-latitude river with a complex rainfall-runoff regime, where a large impact of regional climate warming and glacier retreat are strongly manifested, The Athabasca River Basin was selected for this study. Development of Nutrient and Pesticide Concentrations in the Sázavka River and Their Changes in Connection with the Rainfall-runoff Regime

This bachelor thesis aims to study the selected parameters and nutrients of water quality in relation to the rainfall-runoff regime in the Sázavka River basin. The rainfall-runoff regime is influenced by many factors, including the climate change, which has received more and more attention in recent years. These changes may have a consequential impact on the quality of surface waters. In the first part of this paper selected parameters of water quality are presented. The aim was to try to understand their behaviour under the changing water flow dynamics. For this purpose, Czech and foreign literature was used. In this thesis there are also presented the most common statistical methods that are used in analysing geochemical parameters. The second part of this paper deals with the changes of climate- hydrological characteristics (air temperature, precipitation, snow cover and flow), the analysis of nutrients (forms of nitrogen and phosphorus) and physical parameters (water temperature and electrolytic conductivity). Lastly, metabolites of herbicides that ware above the detection limit in the watershed (acetochlor ESA, alachlor ESA, dimethachlor ESA and metolachlor ESA) were assessed. nutrient supply in vegetarians

The bachelor's thesis deals with the supply of nutrients in ovo-lacto vegetarians. The goal of the work is to find out whether the supporters of this form of vegetarian diet are sufficiently supplied with macronutrients and selected micronutrients, the main area I am targeting in the research part of the work. The theoretical part is first focused on vegetarianism as an alternative way of eating, its definition, history in the world and in the Czech Republic, reasons for adopting vegetarian forms of eating and analysis of individual forms of vegetarian diet. The second section of the theoretical part looks at the advantages and disadvantages of the ovo-lacto vegetarian diet and the analysis of individual nutrients. Mainly the nutrients that are part of the research are analyzed, the vast majority of which are those nutrients that can be contained in a vegetarian diet in lower concentrations than in a mixed diet. Twelve vegetarian men and women aged 20-41 were included in the research investigation of the practical part of the bachelor's thesis. First, the respondents' diet records in terms of intake of individual nutrients were evaluated in the Nutriservis application, and the resulting average concentration of nutrients in the diet was then compared with the recommended values for nutrient intake. In addition to a separate evaluation of the menus of individual respondents, specific macronutrients and micronutrients were subsequently analyzed, or more precisely, their average occurrence in the menus of all respondents. The results of the research point to deficiencies in the quality and, above all, the quantity of macronutrients and selected micronutrients in the respondents' diet.

The diploma thesis focuses on the analysis of the nutritional level of dairy cows and its influence on selected qualitative indicators of milk production. The aim of the work is to carry out a comprehensive assessment of the impact of feed ration and its composition on the quality and quantity of milk production, with an emphasis on identifying optimal nutritional conditions for achieving the highest utility and ensuring good health and living conditions for dairy cows. In the first part of the work, a theoretical review of current knowledge about dairy cow nutrition and milk quality was carried out, focusing on energy and nutrient needs of dairy cows, quality and optimal representation of feed, individual phases of lactation, milk quality, microbiological and hygienic value of milk, and factors affecting its quality. In the second part of the work, an analysis of the nutritional level in the selected breeding was performed. Mixed feed rations and individual components contained in them were evaluated here. Furthermore, utility and qualitative indicators of milk (fat content, protein, lactose, somatic cells, and total number of microorganisms) were assessed.

This bachelor thesis aimed to characterize the effect of imidacloprid (neonicotinoid insecticide) on nutrient levels (carbohydrates, proteins, lipids) in haemolymph, on the activity of digestive enzymes (amylases, proteases, lipases) in the gut, and on adipokinetic hormone and biogenic amine levels in the central nervous system of the worker honeybees Apis mellifera. The insecticide was applied topically to the bee body at a dose of 0,04 micrograms per bee. The above-mentioned characteristics were measured 24 hours after insecticide application. The results showed that the effect of imidacloprid on the nutrient level in haemolymph was variable - the level of carbohydrates and proteins decreased, while the level of lipids increased. The activity of digestive enzymes in the gut increased (proteases, lipases) after the application of imidacloprid. On the other hand, the levels of adipokinetic hormone and biogenic amines in the central nervous system were reduced.
Food supplements in strength sports

In this bachelor's thesis on Dietary Supplements in Strength Sports, I investigated whether elite athletes are able to consume sufficient nutrients without dietary supplements and what types of dietary supplements are used by strength athletes. I chose the respondents from three branches of strength sports, those sports are power triathlon, strongman sport and weightlifting. These are the leading athletes in these sectors in the Czech Republic and they are able to outline to me the topic of taking dietary supplements. The theoretical part is focused on the nutrition of athletes. In each section I have discussed a description of strength sports, including the history of strength sports, then I have discussed macronutrients, micronutrients and also the issue of dietary supplements, where I have described the history of dietary supplements and also their distribution. In the theoretical part I drew from books and professional journals. For the practical part of the thesis I set three objectives. The first objective was to map what types of dietary supplements are used by strength athletes, the second objective was to map what amounts of dietary supplements are used by strength athletes, and the third objective was to determine if a regular and complete diet provides the necessary amount of energy and individual nutrients. Ten athletes participated in my research, 9 men and 1 woman. I obtained data from them through a semi-structured interview and from their dietary records. The results show that the respondents are taking 10 types of supplements and 6 out of 10 respondents are able to take sufficient energy from their regular diet. The dietary analysis shows that respondents prefer a diet higher in fat and protein at the expense of carbohydrates, where a higher proportion of fat in the diet reduces the amount of food needed to meet the respondents' energy needs. It can also be observed that respondents do not place much emphasis on fibre intake.

The thesis gives an overview of the nutrition of calves and heifers. The thesis is focused mostly on the dairy breeds of cattle and contains recommendations about the correct technology of rearing of calves and heifers, composition of rations, feeding in individual periods with regard to the anatomical-morphological structure of the individual organs of an individual which changes with age. The last chapter is made up of the problematic of health. Heifers are a category of cattle from 6 months to calving. It therefore follows the plant nutrition of calves and is exclusively based on plant feeds. Suitable feed ration in necessary for good health which is a prerequisite for high production of good-quality product and ensuring longevity of dairy cows. The basic requirement for feed ration is to match the growth intensity set by the breeding goal. The correct rearing of heifers is an important factor for high production and correct reproduction.
